A2A adenosine receptor interacting partners (interactome)

چکیده
Background The A2A adenosine receptor is a member of the G proteincoupled receptor family. It is known to posses several unique structural and functional features that differ from established signaling paradigms. Recently, the exceptionally long (>120 amino acids) C-terminus of the receptor has raised interest for its capacity to act as a binding site for several "accessory" proteins. In search for interaction partners we identified a list of possible candidates via a yeast two-hybrid system. From the identified potential interaction partners we further investigated (i) ARNO/ cytohesin-2, a guanine nucleotide exchange factor for the small G protein ARF6, and (ii) SAP102, ''synapse-associated protein of 102 kDa'', and their effects on the signaling properties of the A2A receptor.

The interactome of the A2A adenosine receptor in vitro and in vivo
Background The A2A adenosine receptor is a prototypical G proteincoupled receptor. It is expressed in a wide variety of cells including as different types as neurons, platelets, cells of the immune system and muscle. The cell-specific expression of the A2A adenosine receptor is controlled by at least three different upstream non-coding exons and their corresponding promotors. Compared to other ...

Background The A2A adenosine receptor is a prototypical G proteincoupled receptor. It is expressed in a wide variety of cells including as different types as neurons, platelets, cells of the immune system and muscle. The A2A receptor has an unusually long C-terminus (of >120 residues), which for the most part is dispensable for coupling to Gs. This C-terminus turned out to be the docking site f...

Objective(s): For determining the mechanism of anti-asthmatic effect of thymoquinone, this investigation evaluated the effect of thymoquinone in the presence of selective A2A and A2B adenosine receptor antagonists (ZM241385 and MRS1706, respectively). Materials and Methods: Seventy guinea pigs were randomly divided to 7 groups; control (C), sensitized with ovalbumin (S), sensitized groups pretr...
متن کاملEffects of adenosine A2a receptor agonist and antagonist on cere-bellar nuclear factor-kBexpression preceded by MDMA toxicity
Background :Adenosine is an endogenous purine nucleoside that has a neuromodulatory role in the central nervous system. The amphetamine derivative (±)-3,4-methylenedioxymethamphetamine (MDMA or ecstasy) is a synthetic amphetamine analogue used recreationally to obtain an enhanced affiliated emotional response. MDMA is a potent monoaminergic neurotoxin with the potential of damage to brain neu...
متن کاملP138: Are Depression and Anxiety Affected by Adenosine A2A Receptors?
Adenosine acts as neuromodulator in the brain, which its involvement in a wide range of brain processes and diseases has been studied, such as epilepsy, sleep, anxiety, panic disorder, Alzheimer’s disease, Parkinson’s disease and schizophrenia. Adenosine receptors have been detected: A1R, A2AR (A2AR), A2BR, and A3R. A1R and A2R inhibit cAMP production, while A2AR and A2BR stimulate cAMP product...
